You should be able to test this easily enough. It will only work with a held weapon, so grab a cestus, punch without 1605, punch after 1605 - should show up if there's any difference. It will not be able to apply to kick DF because there is no weapon involved.

If you mean with unarmed combat attack commands (jab, grapple, punch) the answer is no. The 1605 DF bonus only applies to the approved-for-use unarmed combat brawling weapons when they are used in melee combat with the ATTACK and AMBUSH commands. There is also no 1605 DF bonus with jab, grapple and punch attacks when empty handed.
